Screening of target genes for the drug histamine.

To explore histamine target genes,we used CRISPR-PoolTMKOUT-Human-Single vector to build GeCKO(genome-scale CRISPR-Cas9 knockout) human library.Then Genome-wide CRISPR screen was used to detect target genes in cholangiocarcinoma cells. Overall design: After GeCKO library was successfully constructed using lentivirus in human cholangiocarcinoma cell lines, the nuclear genome was extracted for PCR(Polymerase Chain Reaction) purification and amplification, then the PCR amplification products were detected by Genome-wide CRISPR screen.
