Species-dependent in vivo mRNA delivery and cellular response to nanoparticles

Delivery of LNPs in humanized mice to determine transcriptional differences in human cells. Overall design: Humanized mice were administered ether pooled LNP libraries or PBS as a control using IV injection. Livers were digested with Collagenase and hepatocytes were sorted with FACS. In JD69 samples 3 mice per group were pooled prior to RNA-seq library prep. For the remaining samples mice were kept separate for biological replicates. Library prep was performed using modified SMART-seq protocol for 1000 cells.
