Targeted bisulfite sequencing of selected CpGs within GR-binding regions of the ZBTB16 locus

Human cerebral organoids were treated with 100nM of dexamethasone or equal volume DMSO for 7 days starting at day 43 of differentiation. The methylation landscape of the ZBTB16 locus was analysed using targeted bisulfite sequencing at vehicle- or in response to dexamethasone treatment. We focused on intragenic regulatory elements that encompass GR-binding sites. We found altered methylation levels of CpGs located in immediate regions next to or in GR-binding sites in response to dexamethasone treatment as compared to vehicle and/or to CpGs in non-GR binding regions.
