To identify and characterize transcript structures ranging from the transcriptional start sites (TSSs) to the termination sites (TTSs), we constructed and analyzed TSS/TTS mate pair full-length cDNA libraries from 18 kinds of human cell types. Furthermore, we applied the similar strategy to construct cDNA libraries in which 3’-end mate pairs distribute randomly over the transcripts. We show the TSS/Random libraries were useful to assembly internal structure of the previously uncharacterized alternative promoter products as well as intergenic transcripts. This novel method to analyze full-length cDNAs should be useful to complement currently RNA Seq analyses and genome annotations.
