TNF-a is elevated in a variety of inflammatory disease involving epithelial tissues. To better understand the the impact of TNF-a on epithelial biology, 3 epithelial cell lines were cultured with and without TNFa and the transcriptomes compared using were profiled using Affymetrix GeneChip® HG-U133+2 array or HG-U133+PM arrays. Overall design: Three epithelial cell lines (A549, HT-29, 293) and K562 (mylegenous leukemia) cell line were cultured in 6 replicates for 4 hrs with and without TNF-a and transcriptomes were assessed using Affymetrix GeneChip HT HG-U133+ PM 96 array plates. A list of genes from TNF-stimulated cultured primary human keratinocytes were also obtained for the experiment (original historical dataset not available). Data from K562 were not included in the summary gene set for TNF stimulation.
