Hematopoiesis generates a variety of cell types after M3434 culture. We treated HSPCs with small-molecule inhibitors of Acly (SB204990), Gls (CB-839), or Vehicle (DMSO) and compared them to freshly isolated HSPCs by single-cell RNA-seq and single-cell ATAC-seq to examine the diverse cell types emerging from these conditions. Overall design: HSPCs were isolated by Lineage-depletion and c-Kit enrichment by MACS cell sorting and either cultured for 2 weeks in M3434 or isolated fresh from BL/6 mice and analyzed using scRNAseq or scATAC-seq.
