The soil microbial biomass prototype study was carried out in order to evaluate the methods commonly used for measuring microbial biomass. A field sampling campaign was carried out to collect soil samples and analyze samples using chloroform fumigation, substrate-induced respiration (SIR), traditional and high-throughput phospholipid fatty acid (PLFA) analysis, quantitative PCR (qPCR), and direct counts. This study was conducted at the NEON Soaproot (SOAP) and San Joaquin Experimental Range (SJER) sites, which are located in NEON Domain 17.
