Chromosome DNA replication is a fundamental process of life, involving the assembly of complex machinery and dynamic regulation. Here, we reconstruct a bacterial replisome module by artificially clustering 23 genes that participate in DNA replication and sequential deleting these genes from their natural scattered chromosome loci in Escherichia coli.
