The transcription factor PU.1 plays a crucial role in macrophage development. We examined the role of PU.1 in macrophage activation induced by a bacterial cell wall component lipopolysaccharide (LPS) and interleukin 4 (IL4) by using the PU.1 selective inhibitor DB2313 in bone marrow-derived macrophages. Macrophages were pre-treated with DB2313 (500 nM) for 18-24 h and then activated by LPS (100 ng/ml for 5 h) or IL4 (20 ng/ml for 18 h). Total RNAs were harvested and sequenced by an Illumina sequencer.
