Cofilin1-driven cytoskeletal remodeling licenses pan-cancer CD8+ T cell immunotherapy responses

This dataset contains processed single-cell RNA sequencing (scRNA-seq) data from 8 patients with urothelial carcinoma (UC) who received neoadjuvant tislelizumab (anti-PD-1 antibody) monotherapy prior to radical cystectomy. The study aimed to identify molecular determinants of immunotherapy response in solid tumors, with particular focus on tumor-intrinsic biomechanical regulators of CD8⁺ T cell-mediated cytotoxicity. Among the 8 patients, 3 achieved pathological response (responders, R) and 5 showed no significant response (non-responders, NR). Tumor biopsies were collected prior to treatment initiation and processed for single-cell transcriptomic profiling.The capture of individual cells and the subsequent synthesis of complementary DNA (cDNA) were accomplished utilizing the Single Cell 3′ Library and Gel Bead Kit V2 (10X Genomics, catalog number 120267).
