As PRC2 shows preference for binding to unmethylated CG-rich sequences in mammals, we asked whether the observed H3K27me3 remodelling is due to recruitment of PRC2 in response to global DNA demethylation.To understand the potential relationship, we categorized promoters based on the changes in DNA methylation and H3K27me3 between E10.5 and E13.5 (see Methods). We observed four patterns: Group A, loss of 5mC, gain of H3K27me3; Group B, loss of 5mC, continuous H3K27me3 enrichment; Group C, devoid of 5mC, continuous high level of H3K27me3; Group D, loss of 5mC, devoid of H3K27me3.
