Differential Expression Profiles of tRNA-derived Small RNAs in Muscle-Invasive Bladder Cancer in Chinese Population

The goals of this study are to screen differential expression profiles of tRNA-Derived small RNAs (tsRNAs) in muscle-invasive bladder cancer (MIBC) in Chinese population using next-generation sequencing and qRT-PCR.As a result, in MIBC tissues, 406 tsRNAs were differentially expressed between in MIBC tissues. Of them, 91 tsRNAs were significantly differentially expressed (fold change>1.5; P<0.05): 65 tsRNAs were significantly up-regulated whereas 26 were significantly down-regulated in MIBC.Taken together,this study explored, for the first time, the significant alteration of tsRNA expression profiles in MIBC in Chinese population and offered deep insights into many possible treatment targets of MIBC by regulating tsRNAs. Overall design: This sequencing study includes tissue samples from 4 MIBC patients, all of whom had undergone surgery for MIBC at the department of urology in Beijing Friendship Hospital affiliated to Capital Medical University (Beijing, China) in 2021. This study was approved by the ethics committee of the Beijing Friendship Hospital affiliated to Capital Medical University and conducted according to 1975 Declaration of Helsinki ethical standards, as revised in 2008. MIBC specimens (T group) and adjacent control mucosal tissues (N group) with a distance of 3 cm from the tumor) were collected during the surgery and immediately stored in liquid nitrogen to prevent RNA degradation. In total, 4 pairs of tissues (tumor and paracancer) were used for tsRNA sequencingAll patients who participated provided written informed consents.The expression profiles of tsRNAs were generated by deep sequencing, using NextSeq 500/550 V2 kit (#FC-404-2005, Illumina).
