We created a mutagenesis library to 25 native E. coli sequences known as the "promoter islands." We measured the mutant DNA sequences for promoter activity using a dual reporter plasmid and fluorescence-activated cells sorting (FACS). Sequences are binned into different replicates and fluorescence levels.
