Enzymatic Methyl-seq (EM-seq) is an enzyme-based method giving us reliable methylome data from small amounts of purified DNA. However, it is unclear whether EM-seq library can be constructed from crude cell lysate containing genomic DNA. We evaluated the quality of EM-seq libraries directly prepared from crude cell lysate. Overall design: We prepared the crude cell lysate containing genomic DNA from human induced pluripotent stem cells (iPSCs). We then constructed the EM-seq libraries using crude cell lysate that contained genomic DNA of 10, 50, and 200 ng (crude-10ng, crude-50ng, and crude-200ng, respectively). We also constructed the libraries using purified genomic DNA of 10, 50, and 200 ng inputs (purified-10ng, purified-50ng, and purified-200ng, respectively).
