Transcriptome analysis of regional idiopathic pulmonary fibrosis airway epithelial cultures

We performed RNA sequencing of idiopathic pulmonary fibrosis (IPF) epithelia obtained from different regions of the disease lung and cultured at air-liquid interface. We analyzed cultures from large airways (>10 mm), small airways (<2 mm), non-fibrotic distal airways, fibrotic distal airways, and honeycombed regions. Epithelial cells were isolated individual IPF donor lungs from each region and cultured at air-liquid interface. RNA was isolated from cultures at various timepoints. Illumina TruSeq library prep was performed on 500 ng from each sample (1 per donor, per timepoint) and ran on the NovaSEQ 6000 with 80 million reads per sample.
