The study aimed to understand the characteristics of HSC-NK cells differentiated from HSPCs under hyperosmosis. Through RNA sequencing analysis, the researchers compared differentiated HSC-NK cells under different osmotic pressures and identified minimal changes in the transcriptome due to hyperosmosis. However, expanded NK cells exhibited distinct gene expression profiles compared to uncultured NK cells, indicating activation. Gene Ontology analysis revealed upregulated signaling pathways associated with NK cell activation and immune responses. Overall, hyperosmosis facilitated efficient differentiation and expansion of HSC-NK cells without significant transcriptomic alterations, while promoting an activated gene expression profile in the expanded NK cells.
