This RNA-seq dataset is from RNA sample collected using the physical seperation technique previously used in our lab (Lovelace 2018). The goal is to look at DC3000 AlgU regulon under pattern triggered immunity. Overall design: There are six samples in triplicate for a total of 18 samples. Two mutant strains of P. syringae pto DC3000 were used: ?algUmucAB (GS_00585 “3d“), and ?algUmucAB + AlgU complemented via pEM53 (pBAD promoter) plasmid (GS_00587 “3dA”). 5-week old A. thaliana were pre-treated 20hr in advance of inoculation with either Flg22 (“F”) or mock (water+DMSO, “M”). Inoculum was prepared in 0.25mM MgCl2 from KB agar plates. Pellets (P) of both inoculums were saved as time-zero baseline examples. After 5hrs of infection, plants were vacuum-infiltrated with an RNA-later-like buffer (“M” or “F”) and were frozen in liquid nitrogen until RNA extraction.
