PBMCs were isolated from 4 AH patients and 4 age-matched healthy controls. Cells were cryopreserved, then thawed for ex vivo culturing. Cells were challenged with 100pg/mL LPS (low-dose LPS) for 24 hours to model low-level endotoxemia observed during alcohol consumption. Cells were then harvested and 10x single-cell RNA-seq was performed.
