At the top, the mutation strategy compared to the wild-type allele of the TRIM5 gene. The mutation strategy consists in 8 introduced substitutions, including 2 non-silent substitutions to mutate Arg332 into Gly (blue), 1 non-silent substitution to mutate Arg335 into Gly (blue), a silent mutation to disrupt the PAM sequence (purple), a silent mutation to create a HaeIII restriction site (green) and 3 silent mutations of the sequence targeted by gRNA1 (grey). The HDR-modified region was amplified by PCR and the PCR products were then analyzed by Illumina Miseq sequencing.
