Region-specific alteration of histone modification by LSD1 inhibitor conjugated with pyrrole-imidazole polyamide

We conjugated NCD38 with two different Py-Im polyamides and analyzed the regions epigenetically altered by parental NCD38 and the two conjugates. Human colorectal cancer cells, RKO, were treated with conjugated NCD38 with two different Py-Im polyamides (P4_NCD38 and PIPP_NCD38) for 4 days or 30 days followed by total RNA extraction and ChIP (H3K4me2, H3K4me3 and H3K27Ac). For control, RKO cells were treated with Py-Im polyamides(P4 and PIPP) for 30 days followed by ChIP. ChIP-seq and RNA-seq were performed using NexSeq and HiSeq.
