Here we use a transcriptomic approach to investigate the molecular underpinnings of thermal acclimation in the model diatom species Phaeodactylum tricornutum by comparing the differential gene expression in cultures acclimated to sub-optimal, optimal, and supra-optimal temperatures (10, 20 and 26.5 °C, respectively). Overall design: Cultures (n=4) of Phaeodactylum tricornutum were grown for more than 10 generations at three temperatures (10, 20 and 26.5 °C). Cells were harvested in exponential growth phase by filtering at least 50 mL on to a polycarbonate membrane and flash freezing in liquid nitrogen. Cultures grown at 20 °C are used as a reference transcriptome.
