We employed scRNA-seq to generate a cell atlas of the early leaf development in the wild model grass Brachypodium distachyon. Samples were collected from 15 leaf developmental zones (per dataset) (2021, 2022 and 2024 datasets) and 8 or 10 vegetative shoot apices with young leaf primordia (2023 datasets) of 3 weeks old soil-grown plants. Two genotypes were harvested: Wild type Bd21-3 (WT) and the sid/bdmute-1 (sid) mutant. The tissue was digested to release single cells (protoplasts) which were then sent for 10X Genomics single-cell sequencing. The 2022 libraries were sequenced twice (indicated by -1 and -2).
