The extracellular vesicles (EVs) have an important role in intercellular communication as they transport a rich cargo of organic molecules such as lipids, nucleic acids, and proteins. Different aspects may influence EVs loading, as cell source and the environmental culture conditions. EVs derived from stem and progenitor cells, as expanded CD133+ cells (E-CD133) from human umbilical cord blood (hUCB), have pro-regenerative and pro-angiogenic potential, as we have previously verified by their proteic content. To provide a wider spectrum of the potential cargo of these membranous particles, we now identified the small non-coding RNA content from E-CD133-EVs produced in both normoxic (21% O2) and hypoxic (5% O2) environment, which may influence EVs production and cargo. As an external control, we used EVs from human primary dermal fibroblasts (HDF) cultivated in the same conditions; and sequenced the small non-coding RNA from both HDF and E-CD133 cells. The dataset provides information regarding the differential loading of EVs produced in both normoxia and hypoxia and the potential content of these particles for different applications, as for regenerative medicine.
