Starting from frozen PBMCs of a patient with T-bet deficiency and a wild type control, CD4+ cells were enriched by magnetic sorting. The cells were either activated with plate-bound anti-CD3/anti-CD28 monoclonal antibodies for 14 hours or left untreated. Single-cell RNA sequencing (scRNA-Seq) was performed to study differentially expressed genes in the T helper cell compartment in T-bet deficiency.
