Endocycle is an alternative cell cycle during which the DNA is replicated in the absence of cytokinesis, resulting in cellular endopolyploidy. The endocycle is frequenctly observed in plant species that grow under extreme conditions. Thus, endopolyploidy has been postulated to be a mechanism facilitating adaptive growth. We used microarrays to assess endoploidy-dependant gene expression profiles in Arabidopsis thaliana root and to understand how subpopulations of cells with different endoploidy contributes to plant growth under different environmental conditions Arabidopsis thaliana (pCO2:YFP-H2B , Col-0 ecotype) root cortical nucei were sorted by FACS based on YFP fluorescence and DNA content (2C, 4C, 8C and 16C), subjected to RNA extraction and Affymetrix ATH1 array hybridisation. Prior to hybridisation two consecutive rounds of RNA amplifications were performed using standard Affymetrix protocols for small samples. The amplified nuclear RNA of each DNA content class was used for microarray analysis.
