To investigate the role of salicylic acid biosynthesis in transcriptional response to cytoskeleton disruption, we treated WT and sid2 plants (mutant deficient in ICS1) by latrunculin B for 24 h. We then performed gene expression profiling analysis using data obtained from RNA-seq of 3 biological replicates. Fully expanded mature rosette leaves (three per plant) of Col-0 or sid2, were treated by infiltration of 1 µM of latrunculin B (Sigma) or 0.01% DMSO (Sigma) by needleless syringe. After 24h, leaves were detached and were fixed in liquid nitrogen and total RNA was extracted using RNAeasy Mini kit (Qiagen).
