Melatonin is a powerful anti-inflammatory agent and antioxidant. To explore specific anti-inflammatory mechanisms, LPS was used to intervene in RAW 264.7 cells to construct an inflammation model. Then, cells were pretreated with melatonin at concentrations of 10-3M to 10-7M, respectively. The above cells were subjected to DRUG-seq sequencing to explore the specific mechanism of melatonin anti-inflammation. Overall design: To investigate the specific mechanism of melatonin anti-inflammation, RAW 264.7 cells were divided into four groups: control, LPS, melatonin and LPS+melatonin groups. Among them, the control group was intervened with solvent only. the LPS group was treated with 10µg/mL of LPS. In order to investigate the concentration effect of melatonin, the melatonin group and LPS+melatonin group could be divided into five subgroups. Among them, the melatonin group was intervened with melatonin at concentrations of 10-7 M, 10-6 M, 10-5 M, 10-4 M, and 10-3 M. The RAW 264.7 cells in the LPS+melatonin group were pre-treated with a concentration gradient of melatonin for 30 min before the addition of LPS. the above cells were treated for 12 hrs and then collected for the next sequencing step.
