In this study, we used mass spectrometry–based proteomics to analyze renal tissues from normal and DKD mice, with validation by qRT-PCR and Western blot. Our findings aim to provide mechanistic insights and potential therapeutic targets for DKD.DKD mice exhibited hyperglycemia, insulin resistance, dyslipidemia, and renal dysfunction. Electron microscopy showed glomerular basement membrane thickening and podocyte foot process effacement. Proteomics revealed 2,950 differential proteins enriched in autophagy and mTOR signaling pathways. Compared with controls, PRMT1, mTOR, and P-Akt/Akt were upregulated, while NPRL2, Beclin1, ATG12, ATG7, and LC3-II/LC3-I were downregulated in DKD kidneys (P<0.05).
