Low-dose carboplatin chemotherapy can enhance CAR T cell potency, resulting in tumor eradication in tumor bearing mice. Mechanistically, carboplatin induces tumor cell death and pro-inflammatory modification of the tumor microenvironment that overcomes immune suppression, resulting in early, intense and persistent infiltration of activated CAR T cells into the tumor. Patients with carboplatin-sensitive prostate tumors may benefit from low dose carboplatin prior to LeY CAR T therapy.
