We performed high-throughput snRNA-seq and spatial transcriptomic RNA-seq on dorsolateral prefrontal cortex (BA9) tissue in Macaca fascicularis to identify cell-type specific differentially expressed genes. The macaques include depressive-like, healthy control and resilient individuals. Overall design: 10X Genomics Chromium snRNA-seq was performed on nuclei extracted from dlpfc of the post-mortem brain tissue of 7 depressive-like (DL), 5 mild depressive-like in low rank (resilient, RES) and 7 healthy (HC) female cynomolgus macaque (Macaca fascicularis). 10X Genomics Visium Spatial transcriptomic was performed on tissue slices from dlpfc of the post-mortem brain of 2 depressive-like (DL), 3 mild depressive-like in low rank (resilient, RES) and 3 healthy (HC) female cynomolgus macaque. Cell-types were identified by unsupervised clustering and diffrential gene expression was performed between cases and controls for each cell-type cluster. The library preparation was performed with the Chromium Singel Cell 3' Reagent Kits v2. Paired-end sequencing was performed on theIllumina NovaSeq 6000. FastQ files were produced with Cellranger v2.1.0 mkfastq. Alignments and gene counts were generated with Cellranger v2.1.0 count.
