RAW 264.7 RNA sequencing of fever at discrete metabolic timepoints

A global RNA sequencing of combined LPS-and-IFNg-stimulated mouse macrophage cell line (RAW 264.7) at baseline temperature, 37C, and fever temperature, 39C. Incubated monocultures were harvested at distinct timepoints based on a 400-minute metabolic kinetics assay. Point "B" measured OCR and PER at basal conditions prior to stimulation and point "A" measured macrophage activation 25 minutes post-stimulation. A later metabolic reprogramming occurs in which OXPHOS rates decreased due to NO production while glycolysis rates increased to compensate for the metabolic deficit. Timepoints "N" and "G" represent these events respectively.
