Endophytes, microbes residing within plant tissues, carry functional genes that complement host-plant needs and support its health, fitness, and adaptation to changing environmental conditions. Understanding which factors influence seed microbiome assembly is vital for comprehending plant-microbe interactions and ecosystem functioning. in this study, the model plant Setaria viridis was grown in different soil types with contrasting microbial richness (Negev and PotMix), and the effect of soil type and microbial richness on the composition of seed-associated prokaryotic communities were determined by applying a high-throughput sequencing-based protocol that targets PCR-generated amplicons from V4 variable regions of the 16S rRNA gene using the bacterial primer set 515F and 806R
