Identification of potent antibodies in human serum is difficult due to limited B cells targeting specific antigens. Our efficient method enriches high-affinity antibodies against pathogens, even without knowledge on immunogens structure. Our two-step screening approach allows the selection selects high-affinity therapeutic human antibodies, in particular against SARS-CoV-2 spike protein, within weeks.
