High-throughput sequencing of 3T3-L1 cells with Kdm5c knockdown

One day prior to confluency, non-targeting (scramble control) siRNA or Kdm5c siRNA were transfected into 3T3-L1 cells and gene expression was assessed by RNA-seq at 24hr, 48hr, and 72hr post transfection. 3T3-L1 preadipocytes were reverse-transfected with 50 nM non-targeting (scramble control) or Kdm5c siRNA with Lipofectamine RNAiMAX at differentiation day –3 (sub-confluent). For RNA isolation (transcriptome analysis), cells were collected at 24hr (day –2), 48hr (day –1), and 72hr (day 0) post-transfection.
