4D-STEM tilt-series of mitochondria in U2OS cell

The details of three mitochondria are clearly visible in the field of view of 37 tilt-view projections. Each projection was acquired in scanning-TEM mode on T-F20 200 keV with 10um overfocused 1.9 mrad electron beam, using a raster scan of 1024x1024 beam positions at 3nm steps. Dectris Arina 96x96 images captured the diffraction plane showing a BF disc diameter of 53 pixels. The tilt angles were according to the rawtlt file in dose symmetric order, starting with s0 files at 0 deg, then s1 files at -3 deg, etc. Scanning and synchronization are based on a SerialEM server "Savvyscan" implemented on FPGA/Linux card Red Pitaya STEMlab 125-14. Processing was based on the shadow montage workflow (https://github.com/Pr4Et/Supplementaries/tree/main/Shadow_Montage), specifically ShadowMontage_tiltseries_ver2.m with Ns=4. Alignment was carried out using IMOD.
