Nucleo-cytoplasmic distribution of SAP18 reveals its dual function in splicing regulation and heat-stress response in Arabidopsis

RNA-seq experiment of Col0, sr45, sap18sr45 and asap (sap18sr45acinus) mutants Col0, sr45, sap18 sr45 and asap plants were grown on MS medium under 16:8 photoperiod at 22 °C for 11 days. RNA was extracted from 100 mg of seedlings using RNeasy plant mini kit (Qiagen) following manufacturer’s instructions. RNA was quantified in a Qubit 2.0 fluorometer using Qubit RNA Assay Kit and following by an integrity check on Bioanalyzer. 2 μg of RNA per sample was sent to Macrogen for library preparation and sequencing performing (Truseq stranded mRNA Library + NovaSeq6000, 150 x 2bp, 30M reads/sample, 4.5Gb/sample).
