Sleep bruxism (SB) is a disorder in which patients repeatedly make involuntary jaw movements during sleep. In this study, we performed single nucleus RNA sequencing (CTRL:SB = 1:1) using iPS cell-derived neurons established from SB patients to investigate gene expression of each neural subtype specific to SB patients. Overall design: To investigate gene expression of each neural subtype specific to SB patients, we established human iPSCs(hiPS) from three SB patients (SB1, SB2, and SB3) diagnosed with SB using clinical diagnostic criteria and also had the risk allele (C) of HTR2A SNP rs6313 and three unaffected controls (CTRL1, CTRL2, and CTRL3) without the risk allele. Neurons were induced to differentiate from each sample. We then performed gene expression profiling analysis using data obtained from single nucleus RNA-seq (snRNA-seq) of two different samples. Comparative gene expression profiling analysis of snRNA-seq data of neurons from patients with SB (SB2) and controls (CTRL2).
