The Venus Flytrap 'DYSCALCULIA' (DYSC) naturally occurring mutant / cultivar refers to the originally called 'ERROR' cultivar discovered by Mathias Maier in 2011 (Bailey & McPherson, Dionaea, pg. 214, 2013). This cultivar is not able to shut its traps after touching the trigger hairs two times, as a normal wild type (WT) does. To understand the underlying molecular basis of the mutant's disorder we sequenced trap RNA one hour after wounding one trap lobe. The wounding treatment was done by smashing the tissue with a sharp tweezers only one time. During the wounding process, it was important that no trigger hair or the midrib was wounded by the tweezers and to make sure that the tissue was perforated. Wounding for one time elicits three to eight action potentials (AP). Same procedure was done in parallel to the functional WT - which was used as a reference when comparing the two phenotypes. WT traps showed rapid trap closure after wounding. DYSC traps mostly remained open and did not show a fast reaction directly after wounding of the tissue. In addition to the wounded samples, controls - without touch or wounding - were harvested. For each of the different conditions, traps from three biological replicates were used. Single end sequencing was used as RNA Seq Method.
