We have performed single-cell RNA-seq to evaluate whether Gcm2 KO affects chimerism of PTG cell lineages. Overall design: Adult Gcm2-/- mice complemented with mESCs showing 50?70% chimerism of peripheral blood cells were used (three for para10_215 and five for para10_222). Single-cell gel beads-in-emulsion were generated using the Chromium Controller (10x Genomics). Libraries were generated using the Single Cell 3' Reagent Kit v.3.1 for dual index (10x Genomics) according to manufacturer's instructions.
