Bulk 3' RNA-Seq was performed on Embronic Stem Cells (E14) to explore the diverse effect of CTNNB1 mutation alongside GSK3ß inhibition via CHIR99021 serving as a positive control. Overall design: Genomewide expression profile in E14 ESCs with targetted CTNNB1 mutations and GSK3ß inhibition via CHIR99021 for 48 hours. The sequncing libraries contained Read 1 (R1) and Read2(R2) for each sample. Read 1 contained the cDNA information and read 2 only the UMI and sample barcodes. R1 and R2 were combined to build R4 reads file that was alingned to mouse genome build mm10 to calculate the read counts per sample for comparision.
