Single nuclei RNA sequencing (snRNA-seq) dataset characterizing astrocytes and oligodendrocytes isolated from human postmortem APOE e2/3 Alzheimer's disease (AD) and aged-matched non-symptomatic (NS) brains. Overall design: Nuclei were isolated from human post-mortem prefrontal cortex from 16 donors (NS patients = 6, AD patients = 10; all donors were validated as having an APOE e2/3 haplotype). In order to improve astrocyte capture for snRNA-seq, we optimized a platform to enrich for astrocytes by labeling and sorting nuclei based on LIM/Homeobox protein (LHX2)-positive/NeuN-negative staining. Nuclei were then prepared for snRNA-seq using Single Cell 3' Gene Expression kit v3 (10x Chromium). Generated snRNA-seq libraries were sequenced using an Illumina NovaSeq 6000.
