Dataset of loose patch recordings of spontaneous excitatory activity in CA1 pyramidal neurons in a model of chronic epilepsy

This dataset contains the raw data underlying the publication investigating the functional role of Kv7 channels in hippocampal CA1 pyramidal neurons in temporal lobe epilepsy. The study was performed in control tissue and in chronically epileptic tissue from the pilocarpine model of temporal lobe epilepsy. Loose patch clamp recordings were obtained from the perisomatic region of CA1 pyramidal neurons. Putative spontaneous excitatory postsynaptic currents (EPSCs) were induced under conditions of disinhibition and mild extracellular potassium elevation using 5 µM gabazine and 6 mM extracellular K+. The dataset includes recordings used to assess the effects of the selective Kv7 channel blocker XE991 on EPSC frequency, amplitude, and waveform kinetics in control and epileptic tissue. The data support the finding that Kv7 channel blockade affects spontaneous excitatory synaptic activity differently in epileptic versus control CA1 neurons. In epileptic tissue, XE991 produced a stronger increase in EPSC frequency, whereas effects on EPSC waveform duration and amplitude were more pronounced in control tissue. Together with previous evidence of persistent transcriptional downregulation of Kv7.2 and Kv7.3 channels in the pilocarpine model, these data contribute to understanding altered Kv7 channel function and network hyperexcitability in temporal lobe epilepsy. The deposited files are intended to enable data sharing, transparency, and reproducibility of the published analyses.
