Here we have undertaken a gland transcriptome analysis of pre-parasitic J2 and used this data to improve M. chitwoodi annotation and to identify effector candidates relevant for early stages of parasitism. The gland transcriptome shows that pre-parasitic J2s express known and putative novel effectors. We selected a few effectors (both known and novel) and confirmed their gland localization by in situ hybridization, indicating that the gland library transcriptome could functionally replace laborious in situ hybridization assays. By focusing on the gland specific genes, we could identify putative M. chitwoodi effectors. Understanding the complex parasitic mechanisms of Meloidogyne chitwoodi, particularly in its pre-parasitic stage, sheds light on potential targets for nematode control strategies. By identifying and characterizing novel effectors through gland transcriptome analysis, we move closer to developing effective management strategies for this economically damaging nematode species.
