The microRNA cluster miR-106a~363 is expressed from the single X chromosome in male mouse ES cells. The cluster has been targeted by homologous recombination and converted to a conditional alleles. I wish to assess the effect of the targeted deletion and reconstitution of the microRNA loci primarily on the targeted loci themselves. For instance whether the supposed nulls are properly null, and whether reconstitution of the miRNA locus has restored expression correctly.
