To identify early response genes to disulfide stress, cells grown in YES medium to OD600 0.3 were treated with 1.0 mM diamide for 0, 20, 60, 90 and 120 min prior to harvest of total RNA. Poly A RNA from each sample were subjected to fragmentation, size selection, reverse transcription, and adapter ligation. The resulted libraries were 50 bp single end sequenced on a illumina sequencer.
