RNA-sequencing of myelin-specific 2D2 CD4+ T cells isolated from the spinal cord and spleen of mice with EAE. Overall design: CD4+ T cells isolated from the spleen of 2D2 mice were activated by stimulation with aCD3 (1ug/ml) and aCD28 (1ug/ml) in the presence of IL1b (20ng/ml), IL6 (20ng/ml) and IL23 (20ng/ml), aIL4 (5mg/ml) for 96 hours to induce pth17 cell differentiation. Samples of naive and pTh17 were collected. pTh17 cells were transferred into Rag1-/- hosts. Donor 2D2 CD4+ T cells were isolated from the spleen and spinal cord of the Rag1-/- host mice at day 14 after EAE induction, and enriched by FACS.
