ATACseq of human basophils, B cells and CD34-derived basophils

human basophils and B cells (that were purified to >98% homogeneity) and CD34-derived basophils were processed for ATACseq 100,000 to 200,000 resting cells were lysed and treated with the transposition reaction mix (Nextera)(all further preparation used Nextera reagents and tags), fragmented DNA amplified by PCR and fragments run on electrophoresis to isolate 200-1000 bp fragments. Sequencing was performed by the Illumina Nextseq sequencing using high-output 75-cycle reagent kit for 75 bp reads. 100-400 million reads
