Skin inflammation involves complex interactions between immune cells. To address this gap, we applied single-cell RNA sequencing to dissect cellular states and regulatory programs in DPCP-induced dermatitis. This approach enables high-resolution mapping of immune crosstalk and identification of novel macrophage subsets. Our study establishes a framework for understanding how immune and metabolic signals orchestrate skin inflammation and regeneration. Overall design: We isolated CD45+ single cells from DPCP-challenged skin on day 2 and control skin to capture dynamic immune responses. Comparative scRNA-seq profiling across control and inflamed groups was performed to dissect temporal changes in cell states and interactions.
