Proteomic profiling was performed on primary lung fibroblasts from 10 idiopathic pulmonary fibrosis (IPF) patients and 10 control donors. Proteins were extracted using a urea-based lysis buffer, digested with trypsin using a filter-aided sample preparation (FASP) protocol, and analyzed by LC–MS/MS. Data acquisition was conducted in data-dependent mode on an Orbitrap Exploris 480 mass spectrometer. Raw data were processed using Proteome Discoverer™ (v3.1) with the UniProt Homo sapiens database, applying a false discovery rate (FDR) of <1%. Protein quantification was based on precursor ion intensities with total peptide amount normalization.
