We investigated whether LAD disruption phenotypes are associated with transcriptional changes during zygotic genome activation (ZGA). To address this, we performed single-embryo RNA sequencing at the late 2-cell stage and analyzed the transcriptome. Mouse embryos (zygotes) were microinjected with mRNA(s) encoding either mGFP alone (Control) or candidate proteins from the respective pools. The embryos were cultured until the late 2-cell stage and collected for Smart-Seq+5' analysis. Embryos were processed in two batches: batch 1 (collections 1 and 2) included mGFP, Pool J, and Pool M samples, while batch 2 (collections 3 and 4) consisted of Pool I, Pool N, and an additional set of mGFP embryos as controls.
